Fungal Enzymes

Trichoderma reesei cellulase have been extensively studied and characterized, both singly and as mixtures.  C5-6 is proud to offer the only complete set of purified T. reesei cellulose-degrading enzymes as well as purified T. reesei xylanases, α-glucuronidase, β-glucosidase, β-xylosidase, and α-arabinofuranosidase.  All enzymes except CBHI are cloned in Pichia pastoris as secreted proteins.  Proteins are purified by column chromatography to >90% purity and supplied in 50 mM Na-Acetate (pH 5.0), 20% glycerol.   Because CBHI is not expressed in Pichia pastoris, CBHI is purified from T. reesei fermentation broth to >90% purity and supplied in 50 mM Na-Acetate (pH 5.0), 20% glycerol.

*These enzymes are expressed at very low levels in Pichia, and only limited quantities are available.

**GH61 family members are copper-dependent lytic polysaccharide monooxygenases that have been reclassified into family AA9 by CAZy. The enzyme also has a measurable level of hydrolytic activity.

The secreted A. niger alpha-xylosidase has been cloned and characterized and is reported to release xylose from  isoprimeverose and xyloglucan heptasaccarides.

The secreted α-L-fucosidase from the plant pathogenic fungus Fusarium graminearum has been cloned and characterized and is reported to release fucose from xyloglucan and synthetic substrates.  The enzyme may also have applications in removal of fucose residues from mammalian glycoproteins.
