Alicyclobacillus acidocaldarius LAA1

As part of a project in conjunction with the Great Lakes Bioenergy Research Center and the Joint Genome Institute, C5-6 Technologies and Lucigen Corp. isolated, characterized, and sequenced a number of new enzyme-producing aerobic organisms.   Alicyclobacillus acidocaldarius strain LAA1 was isolated in one of the enrichment samples. Alicyclobacillus acidocaldarius was originally named Bacillus acidocaldarius, but was reclassified as a separate genus based on 16S rRNA gene sequence analysis, lipid and fatty acid analysis, phenotypic characterization, and DNA–DNA hybridization experiments.  Alicyclobacillus acidocaldarius strain LAA1 was sequenced by the Joint Genome Institute and assembled into 58 contigs.  We have  cloned the LAA1 β-glucosidase,  produced it in E. coli, then purified it by column chromatography to give >90% purity by SDS-PAGE.  The enzyme is supplied as a solution in 75 mM Tris-HCl, pH 8.0 and 25% glycerol.  The enzyme is thermostable and acid-stable and is compatible with thermostable bacterial and fungal cellulases.  We have also cloned and expressed the LAA1 α-amylase.  This enzyme is also thermostable and acid-stable, and may be useful in starch hydrolysis applications.

Streptomyces flavogriseus  ATCC 33331 

As part of a project in conjunction with the Great Lakes Bioenergy Research Center and the Joint Genome Institute, C5-6 Technologies and Lucigen Corp. isolated, characterized, and sequenced a number of cellulolytic organisms.  Streptomyces flavogriseus  ATCC 33331, since renamed as Streptomyces  pratensis  ATCC 33331, is a cellulose-degrading organism with numerous interesting enzymes.  We have  cloned the Streptomyces flavogriseus  ATCC 33331 xyloglucanase,  produced it in E. coli, then purified it by column chromatography to give >90% purity by SDS-PAGE.  The enzyme is supplied as a solution in 75 mM Tris-HCl, pH 8.0 and 25% glycerol.

Bacillus subtilis expansin

Bacillus subtilis expansin YoaJ (EXLX1) is an unusual protein reported to weaken the structure of cellulose.  The protein has no known enzymatic activity, but is reported to increase the activity of cellulases on filter paper.  We have  cloned the Bacillus subtilis expansin,  produced it in E. coli, then purified it by column chromatography to give >90% purity by SDS-PAGE.  The protein is supplied as a solution in 75 mM Tris-HCl, pH 8.0 and 25% glycerol.
